I have looked at may DDS files, and most are 256x256.
But on some they are 512x512. I was wondering if you could enlarge all of the DDS files to this dimension, and if so if you have to anything to them

it's not really a problem but things will get blurry in-game... don't upsize any skins.
if you just uplarge the skin to a 512x512 skin you won't have to do anything (the uvw are relative, defined between 0-1 rather than 0-255 pixels...)

You can however remake the skins to be that size resulting in much more detail. Although this comes at the cost of game speed as now your graphics card has to deal with textures that are 4 times the normal size.
yes how does it affect speed? is this crucial in any way, compared with for example the poly count?
It all just depends on how good of a system you have. Someone with a good system wont even notice. Someone with a bad system will definitly notice the larger amounts of lag.
Yes, for some of my units i will enlarge them, as my PC can easily handle this sort of stuff, and they will look more impressive.
