As far as I know, space trading sims in the vein of Port Royale don't exist, unless you want some major differences. The best thing I can offer is Freelancer, which is more 3D, but has trading/exploration elements like Port Royale.

Freelancer's trading isn't all that impressive
I would suggest X3-The Reunion. There is a huge economy based off of supply and demand so prices actually change. Furthermore, you can build factories or complexes and sell goods yourselves or just have a whole fleet of manual or automatic traders
