The key to assassination of family members is to train up the assassin to eight plus and then take a shot at them, but do not take repeated shots as the first couple of misses leaves them with security conscious traits and ancillaries and they get VERY hard to kill and very likely to kill your murderer.
But don't be afraid to kill your assassins. They're a weapon just as much as a unit of Triarii. You wouldn't hesitate to send them into a breach to take an enemy capital would you? Don't be afraid of a dead assassin or 6 -- omelettes and eggs my friend.
