    You can´t add new factions and you can´t change the cultures of those factions present either, the only thing you can do is changing the look of it. However, some things are culture-bound, such as the leader and event pics, or the culture penalty. As an example,you could make a second Egypt culture by replacing all, say, Greek cities units with egyptian ones. That that "new" Egypt faction, however, will retain the greek-looking leader and event pictures, unless you change those, too, in which case all Greek cultures will get Egyptian pictures.

    If you have a look around the Scriptorium and its sub-forums you'll find various tutorials about changing faction names, banners, units, buildings etc.

    When reusing a Roman faction, it will start the game allied with the other Roman factions and it will share Line of Sight with them. If the Senate exists it will also get missions from them.

    That reminds me, how do you know which missions the AI Roman factions got? I always thought the Senate missions were for all factions at once, am I wrong there?

    No, the Senate missions are for each Roman faction independently. The first ones are hard-coded (Segesta, Apollonia, Syracuse) and the rest are determined based on the progress made by the faction. To take an extreme example, if you mod the game to start the Julii off in the Steppe, the first mission you might get will be to take the Chersonnees.
