Depending on how many in the first place, anywhere from one to two turns to replenish one. 6 pool max seems to replenish one every turn.

I've sometimes encountered something strange where (mostly after a reload) the recruitment pool would miraculously exceed its limit by one or two units.
Example: as Milan, I had the second level barracks built and third level walls (stone) which should give a pool of 8 units Italian militia in total (6 from barracks/2 from the walls) but after reloading and playing a few turns I would somehow have 9 or 10 units available. This has also happened (infrequently) with ships and playing as other factions, only at cities though. Strange!
If a settlement can recruit a particular unit, and such units are disbanded there, then during the end-turn cycle the disbanded units are added to the recruitment pool, possibly exceeding the maximum size. I've seen a couple of units go up to a max size of 10, but most are between 3-6.
Example, playing Venice, you have a citadel with the max barracks in it, allowing you to recruit Venetian Heavy Infantry with 1 exp, max of 6 units. You let the pool build up to 6, then you recruit 6 units of VHI. Let the pool max out again, disband those 6, and the following turn, your recruitment pool of Venetian Heavy Infantry will be 12. It won't start replenishing until it drops down below 6, but you will have 12 units available for recruitment until you use them.
