There is no option to disband spies, assassins or diplomats. I get rid of them by putting them on a single bireme and heading for the nearest hostile navy. That's the only sure way I've found.

One thing that I think helps determine the success rate of spy missions is the city size.
I've found it harder to sneak into the bigger cities.

I want to add that some traits that family members can pick it will increase the chance in detecting spies and assassins . I don't remember such one but there are bunsh of them .
