I would recommend playing the original MTW/VI first, if only so you can get a feel for the game. There are some noticable differences from RTW/M2TW (especially in regards to the battles), so it would probably be in your best interest to familiarize yourself with the game before anything else.

Since you enjoy the Greco-Roman era of ancient history, you might want to try out Hellenic Total War; it's a total conversion mod that allows you to play in ancient Greece as the Persians and various Greek city-states (Athens, Sparta, etc.). I've only played with that mod a little bit myself, but it looked quite good.
